Impact of membrane protein-lipid interactions on formation of bilayer lipid membranes on SAM-modified gold electrode

چکیده

In protein film electrochemistry for redox-active membrane proteins, the reconstitution of protein-tethered bilayer lipid membranes (ptBLMs) can maximize their functionality and stability at electrolyte/electrode interfaces. To understand impact protein–lipid interactions on ptBLM formation, we tracked process a in presence or absence transmembrane enzyme cytochrome c-dependent nitric oxide reductase (cNOR) mixed self-assembled monolayer (SAM)/Au electrode. Time-resolved surface-enhanced infrared absorption (SEIRA) spectroscopy electrochemical measurements revealed that interaction affected formation kinetics phospholipid electrocatalytic cNOR-modified electrode but not fluidity bilayer.

Theoretical study of protein--lipid interactions in bilayer membranes.

An analysis is given for the perturbation of the order and composition of lipid bilayers near an intrinsic membrane protein. Two cases are examined: the protein influences the lipid order (i.e., "fluidity"), and the protein associates with one component of a lipid mixture preferentially. The order perturbation is studied as a function of temperature and lateral pressure by using Landau--de Genn...

Impact of monoolein on aquaporin1-based supported lipid bilayer membranes

Aquaporin (AQP) based biomimetic membranes have attracted considerable attention for their potential water purification applications. In this paper, AQP1 incorporated biomimetic membranes were prepared and characterized. The morphology and structure of the biomimetic membranes were characterized by in situ atomic force microscopy (AFM), infrared absorption spectroscopy, fluorescence microscopy,...

Engineering Lipid Bilayer Membranes for Protein Studies

Lipid membranes regulate the flow of nutrients and communication signaling between cells and protect the sub-cellular structures. Recent attempts to fabricate artificial systems using nanostructures that mimic the physiological properties of natural lipid bilayer membranes (LBM) fused with transmembrane proteins have helped demonstrate the importance of temperature, pH, ionic strength, adsorpti...

Incorporation of outer membrane protein OmpG in lipid membranes: protein-lipid interactions and beta-barrel orientation.

OmpG is an intermediate size, monomeric, outer membrane protein from Escherichia coli, with n beta = 14 beta-strands. It has a large pore that is amenable to modification by protein engineering. The stoichiometry ( N b = 20) and selectivity ( K r = 0.7-1.2) of lipid-protein interaction with OmpG incorporated in dimyristoyl phosphatidylcholine bilayer membranes was determined with various 14-pos...

Lipid-protein interactions in membranes.

The interactions of lipids with integral and peripheral proteins can be studied in reconstituted and natural membranes using spin label electron spin resonance (ESR) spectroscopy. The ESR spectra reveal a reduction in mobility of the spin-labelled lipid species, and in certain cases evidence is obtained for a partial penetration of the peripheral proteins into the membrane. The latter may be re...
